益肾护骨方对废用性骨质疏松动物模型相关指标影响的研究 被引量:1

摘要 目的:观察益肾护骨方对大鼠的血尿生化水平、股骨生物力学、胫骨骨组织形态计量学指标的影响,探讨益肾护骨方对废用性骨质疏松的改善作用。方法:选取3月龄SD大鼠70只,随机分为正常组、假手术组、模型组、益肾护骨方高、中低剂量组和阳性药物对照组(强骨胶囊),每组10只。模型组、益肾护骨方高、中、低剂量组和阳性药物对照组行右侧坐骨神经切断术,假手术组的手术过程只分离显露坐骨神经后缝合切口。益肾护骨方高、中、低剂量组分别以相当于成人剂量的20倍、10倍、6.25倍灌胃给药(52 g/kg、26 g/kg、16.25 g/kg),阳性药物对照组以0.25 g/kg强骨胶囊灌胃给药,用药4周。检测血尿生化、股骨生物力学、胫骨组织形态计量学指标,行MBP-2免疫组化染色,分析其表达阳性率。结果:血ALP、血BGP、血TRACP、尿HOP/Cr、骨小梁节点数、游离末端数及股骨生物力学指标,模型组与假手术组相比显著升高或降低(P<0.01),予以益肾护骨方干预后,与模型组比较,益肾护骨方高、中、低剂量组相关指标显著降低或升高(P<0.01)。结论:益肾护骨方对废用性骨质疏松大鼠有不同程度的成骨作用,可以不同程度延缓或减轻大鼠废用性骨质疏松的发生。 Objective : To study the effect of Yishen Hugu Decoction on the changes of the blood and urine biochemical levels, the bone structure,bone biomechanical properties in the animal model of disused osteoporosis in rats. Methods : 70 SD rats of 3 months were randomly divided into 7 groups : normal group, sham operation group, model group, high dose of Yishen Hugu Decoc- tion group, medium dose of Yishen Hugu Decoction group,low dose of Yishen Hugu Decoction group and drug control group ( Dry- naria fortunei) ,each group had 10 rats. The rats in the groups of model group,high,medium,low dose of Yishen Hugu Decoction groups and drug control group were given sciatic neurotomy. Normal group was given regular food and water supply, the rats in high, medium,low dose of Yishen Hugu Decoction groups were given gavage by a concentration of 52 g/kg,26 g/kg, 16.25 g/kg dose of Yishen Hugu Decoction respectively, and the rats in drug control group were given gavage by a concentration of 0.25 g/kg dose of Drynaria fortunei. The blood and urine biochemical levels were measured, the femora biomechanical test, the tibia were de- calcified and then stained with HE and Masson stain. Results:The differences of ALP, BGP, Strap ,HOP/Cr, BMD and the numbers of nodes and free ends of model group and sham operation group were statically significant( P 〈 0.01 ). After gavage by Yishen Hugu Decoction, those indexes of high, medium, low dose of Yishen Hugu Decoction groups were obviously higher or lower than that of model group (P 〈 0.01 ). Conclusion.The disused osteoporosis can be prevented effectively by Yishen Hugu Decoction in the index of blood and urine biochemical levels, the bone structure, bone biomechanical properties and disuse osteoporosis in rats.
