摘要
本实验对螺旋藻多糖的抗癌作用进行了体内外实验研究和作用机理探讨。以 0 .3~ 1.5g· L-1对 B3 7乳腺癌细胞的抑制率最高可达 6 8.0 % ,对 K562 白血病细胞抑制率为 4 6 .0 % ,对两细胞的 IC50 分别为 1.4 8和1.56 g· L-1。当螺旋藻多糖以 30 0 ,150和 75mg· kg-1灌胃给药时 ,对小鼠 S180 肉瘤的抑瘤率分别为 55.2 % ,4 4 .6 %和 33.8% ;对 H2 2 小鼠肝癌的抑瘤率分别为 4 7.1% ,36 .4 %和 31.0 % ;对 EAC小鼠腹水癌的抑瘤率分别为 56 .9% ,4 4 .7%和 2 2 .8% ,同时具有明显提高荷瘤小鼠脾指数和胸腺指数的作用。以 MTT法检测螺旋藻多糖的荷瘤小鼠脾淋巴细胞转化和对 NK细胞活性的影响 ,也显示具有提高淋巴细胞转化率和促进 NK细胞杀伤靶细胞的作用。测定螺旋藻多糖对脾淋巴细胞 TNF-α诱生作用 ,大剂量组上清液可达 75× 10 -3 IU· L-1(对照组 34× 10 -3 IU·L-1) ,并有较好的剂量 -效应关系。PSP治疗后的荷瘤小鼠其脾淋巴细胞产生白细胞介素- 2 (r IL- 2 )的功能增强。另外 ,口服螺旋藻多糖还具有一定提升或恢复因使用环磷酰胺所致动物外周血白细胞降低的作用。螺旋藻多糖具有广泛的生物学活性。以往研究多集中于抗衰老 ,提高免疫作用等方面。近年来其在抗肿瘤方面的作用又引起了人们的重视。
The antitumor effects of polysacchrides Spirulina Platensis (PSP) were studied in vivo and vitro.In vitro tests,the highest inhibited rates on B 37 mamma carcinoma cells and K 562 leukemic cells were 68.0% and 46.0% respectively when the concentration was 0.3~1.5g·L 1 .In vivo tests,when the dosage were 300,150,and 75mg·kg 1 p.o,the inhibited rates were 55.2%,44.6% and 33.8%;47.1%,36.4% and 31.0% and 56.9%,44.7% and 22.8% respectively on the transplanted tumor cells of Sarcinoma 180,[HJ5”:4/9]H 22 Hepatic tumor and EAC tumor cells in mice.PSP is biological activated substance which has widely immune effect.It had the effects of increasing the weight of spleen and thymus and of increasing proliferation of activated NK cell activity and TNF α levels from 34×10 3 to 75×10 3 IU·L 1 and of having function of activated splenocyte to produce interleukin 2(rIL 2) in tumor bearing mice.It is suggested that PSP has the ability to inhibit tumor and mechanism is partly to enhance immunity in tumor bearing imce.In addition,enhancing or recovering the number of WBC which reduced by CTX is also important function in treatment of tumor.
