血塞通注射液对局灶性脑梗死大鼠NgR mRNA及蛋白表达的影响 被引量:4

摘要 目的:观察血塞通注射液在不同时点对局灶性脑梗死大鼠Nogo-A受体(NgR)蛋白及mRNA表达的影响,揭示血塞通注射液对脑缺血受损神经元的重塑作用机制。方法:采用改良Zea-Longa方法制备MCAO模型,SD大鼠35只随机等分为假手术组、模型组、血塞通小剂量组、血塞通大剂量组、尼莫地平组。各组动物分别于术后7天、14天、28天麻醉断头取材,行免疫组织化学及荧光定量PCR检测。结果:模型组NgR蛋白及mRNA表达随着缺血时间延长而逐渐升高,到28天时达到高峰,较同时间点假手术组比较均增高(P<0.05);各用药组均比模型组低,术后14天及28天,血塞通大剂量组较模型组显著降低(P<0.05)。结论:NgR参与了脑梗死后突触的再生抑制,血塞通注射液可以抑制NgR蛋白及mRNA表达,从而促进神经再生。 Objective:To observe the effect of Xuesaitong Injection on protein level and mRNA level of nerve growth inhibitory factor Nogo - A receptors (NGR) at different time points after operation. Methods :Healthy mature male SD rats were randomly di- vided into model group, normal saline group, Xuesaitong small-dose group, Xuesaitong high-dose group, positive control group ( ni- modipine group). Each group was raised for 7 days, 14 days and 28 days before being executed, and then the brain tissue was taken out to observe the expression changes of the receptors of NGR by methods of immunohistochemistry and RT - PCR. Results : The expression of NGR in the model group was gradually increased with time prolonging,and reached the high rate on the 28th day controlling with the normal group ( P 〈 0. 05 ). The expression in each treatment group was lower than that in the model group. On the 14th and 28th day,the expression in Xuesaitong high - dose group was significantly decreased compared with that in the model group( P 〈 0. 05 ). Conclusion:NgR is involved in synaptic inhibition of regeneration after cerebral infarction. Xuesaitong In- jection can inhibit the expression of NgR protein and mRNA so as to promote nerve regeneration.
