摘要 建立了高效液相色谱(HPLC)-荧光检测器(FLD)-二极管阵列检测器(DAD)测定调味品中罗丹明B的方法。不同基质样品采用适宜的提取液提取后,以甲醇和0.02mol/L的乙酸铵溶液(70∶30)为流动相,经C18色谱柱分离,用荧光-二极管阵列检测器串联测定,FLD激发波长为547nm,发射波长为573nm,DAD检测波长为550nm,外标法定量。实验结果表明:罗丹明B用FLD检测在1~2000ng/mL浓度范围内呈良好的线性关系,定量限为0.005mg/kg;DAD检测在100~2000ng/mL浓度范围内呈良好的线性关系,定量限为0.5mg/kg。方法回收率在83.1%~105.1%之间,RSD为1.64%~5.42%(n=6)。该方法准确、灵敏,前处理简单,适用于调味品中罗丹明B的定性、定量分析。 A method for the determination of Rhodamine B in condiment by high performance liquid chromatography with diode array detector and fluorescence detector (HPLC-DAD-FLD) is estab- lished. The samples with different matrixes are extracted by suitable extraction solvent, separated by C18 column with methanol and 0.02 mol/L ammonium acetate(70 ~ 30)as mobile phase. The detec- tion is performed by FLD at 547 nm and 573 nm and DAD at 550 nm using external standard method. The experimental results indicate that there's a good linear relationship when Rhodamine B is within the range of 1~2000 ng/mL by FLD, the limit of quantitation is 0. 005 mg/kg. There's a good linear relationship when Rhodamine B is within the range of 100~2000 ng/mL by UV, the limit of quantita- tion is 0.5 mg/kg. The recoveries are 83. 1%~105.1% and the RSDs are 1.64 %-5.42% (n=6). The method is with simple pre-processing, sensitive and accurate, and it can be used for qualitative and quantitative analysis of Rhodamine B in condiment.
