摘要
目的 探讨氧化修饰低密度脂蛋白 (OX- LDL)对血管内皮细胞损伤的机制。方法 在人脐静脉内皮细胞体外培养的基础上 ,通过采用 3H-胸腺嘧啶核苷 (3H- Td R)掺入 DNA技术和硫代巴比妥酸比色法 ,分别分析了 OX- LDL对内皮细胞 DNA合成及脂质过氧化物 (LPO)的代谢产物丙二醛 (MDA)含量的影响。结果 当内皮细胞暴露于 OX- LDL(50~ 2 0 0μg/ ml) 2 4 h后 ,其 3H- Td R掺入明显减少 ,但 MDA含量显著增加。结论 OX- LDL能够明显抑制内皮细胞 DNA合成 ,并显著增加内皮细胞的脂质过氧化物含量。
To study the injury effects and mechanism of oxidized low density lipoprotein(OX LDL) on vascular endothelial cells.Methods On the basis of the establishment of culture medhod for human umblilical vein endothelial cells(HUVECs),the effects of OX LDL on DNA synthesis of cultured HUVECs were observed by 3 H thymidine( 3 H TdR) incorproation and concertrations of malondialde hyde(MDA) of HUVEs were also measured by thlobarbituric acid reaction.Results The smaples of HUVECs were subjected to 50,100,200 μg/ml OX LDL for 24 hour. 3 H TdR incorporation in HUVECs DNA was decreased significantly, and the contents of MDA in HUVECs were increased markedly.Conclusions OX LDL could inhibit DNA synthesis of HUVECs,which might be related to lipid peroxidative injury.
