Study on immunogenicity of microencapsulated newborn porcine islets

摘要 目的 经过体外实验 ,明确微囊化新生猪胰岛样细胞团 (ICC)的免疫原性。方法 通过胰岛—淋巴细胞混合培养 (MLIC)后的淋巴细胞转化试验 ,以游离的新生猪ICC和空微囊为对照 ,比较微囊ICC组与对照组的淋巴细胞转化值。结果 微囊ICC组的淋巴细胞转化值显著低于游离ICC组 (P <0 .0 5 ) ,但明显高于空囊组 (P <0 .0 5 )。结论 微囊包裹后的新生猪ICC的免疫原性比游离的新生猪ICC的免疫原性显著降低 ,提示微囊有良好的免疫隔离作用 ;但微囊包裹后的新生猪ICC的免疫原性相对空囊的免疫原性较高 ,说明免疫隔离作用不完全。 Objective To study the immunogenicity of microencapsulated newborn porcine islet like cell clusters (ICC). Methods After mixed lymphocyte islet coculture (MLIC), lymphocyte transformation test were performed to compare the lymphocyte transformation value (LTV) of microencapsulated ICC (Mc ICC) group with empty microencapsule (empty Mc) group and free ICC group respectively. Results The LTV in the free ICC group (16154±3244)cpm was highest among the three groups. Mc ICC group had a significantly reduced LTV (6989±1561)cpm, P < 0.05 , which was higher than that of empty Mc group (1920±726)cpm, P < 0.05 . Conclusions Free ICC was potent stimulator of lymphoid proliferation and Mc ICC could reduce this stimulation effect significantly. However, the immunogenicity of Mc ICC was still significantly increased as compared with empty Mc.
