胡萝卜软腐欧氏杆菌胡萝卜亚种游动性突变体的筛选

用转座子 Tn5对胡萝卜软腐欧氏杆菌胡萝卜亚种 ( Erwinia carotovora subsp.carotovora,Ecc)进行诱变 ,获得 9个游动性改变了的突变体。 M4 32游动性变大 ;M143、M4 51和 M574游动性完全丧失 ;M4 3、M4 9、M330、M72 5和 M72 6游动性较野生型变小。这 9个突变体在大白菜叶柄上的致病力均减弱。游动性变小或丧失的突变体鞭毛数目减少或未发现有鞭毛。

Nine motility mutants (M43, M49, M143, M330, M432, M451, M574, M725, M726) of Erwinia carotovora subsp. carotovora were obtained by using Tn5 mutagenesis. Mutant M432 exhibited increased motility, showing 40% increment than the wild strain Ecc-12; M143, M451 and M574 were found to be non-motile and non-flagella. While mutants M43, M49, M330, M725 and M726, in which flagella number was decreased,exhibited 40%-50% motility of Ecc-12. The virulence of all mutants on Chinese cabbage reduced to 19%-29% of that in Ecc-12.
