知母中知母皂苷BⅡ含量测定方法的研究

Determination Method of Timosaponin BⅡ in Rhizoma Anemarrhenae

目的:建立HPLC-ELSD法检测知母中知母皂苷BⅡ的含量测定方法。方法:采用Agilent TC-C18(4.6×250 mm,5μm)色谱柱,流动相为乙腈-水(24∶76);漂移管温度为105℃,载气流速为3.0 L/min。结果:知母皂苷BⅡ进样量在0.7850～19.625μg范围内呈良好线性关系,平均回收率为103.1%(n=6,RSD=1.04%)。结论:该方法准确可靠,无干扰,可用于知母药材的质量控制。

Objective ： To establish the quantitative method of timosaponin B II in Rhizoma Anemarrhenae by HPLC - ELSD. Method： The separation was performed on a Agilent TC - C18 （4.6 mm x250 mm,5 μm） packed with a 5 txm sta- tionary phase. The mobile phase consisted of acetonitrile - water （24 ： 76）. The temperature of drift tube was 105 ℃ and the nebulizer nitrogen flow rate was 3.0 L min -i. Results ： The liner range of timosaponin B II was 0.7850 - 19. 625 μg （r=0.9999）. The average recovery was 103.1% （n =6,RSD = 1.04% ）. Conclusion： It was proved that the method with HPLC - ELSD was accurate and could be used for the quality control of Rhizoma Anemarrhenae.
