白头翁和黄连不同配比下有效成分溶出量和量效关系的研究 被引量:1

摘要 目的探讨白头翁与黄连不同配比对有效成分溶出量及对肠运动性能的影响。方法回流提取不同配比的白头翁和黄连,以干浸膏量、白头翁皂苷B4、小檗碱含量为指标,考察白头翁和黄连不同配比对有效成分溶出量的影响;运用肠内容物推进速度实验和离体回肠实验,以小鼠离体肠内容物推进速度和家兔离体回肠震动幅度为评价指标,考察白头翁和黄连不同配比对量效的影响,并分析量效关系。结果经方配比时浸膏和白头翁皂苷B4的得率较其他配比高,但小檗碱的溶出率并不是最高的。肠内容物推进速度实验表明,白头翁-黄连(3∶1)及单味药白头翁与空白组相比有显著性差异;离体回肠法表明白头翁-黄连3∶1、1∶1组和单味药白头翁与空白组相比有显著性差异。结论白头翁汤经方配伍具有较高的有效成分溶出量,并对肠道收缩抑制作用明显。 Objective To explore the dissolution of active ingredients of Pulsatilla and Coptisthe before and after their compatibility, and evaluate the effects of different ratio on intestinal motor function. Methods The different ratio of Pulsatilla and Coptis were selected, after the water boiling reflux extraction, the amount of dry extract, Pulsatilla Saponin B4, berberine volume were obtained for the test, to examine the active ingredients in aspects of dissolution rate changes and compatibility relations; based on the intestinal contents from ileum speed test and ileum experiment in vitro,speed in mice and vibration rate of rabbit ileum were obtained to research the effects of the dose-effect relationship. Results The best dissolution rate of extract and Pulsatilla Saponin B4 was the classical prescriptions, but the dissolution rate of Berberine was not the best in this ratio. Intestinal contents forward speed experiment showed that only Pulsatilla Coptis (3 : 1) and single Pulsatilla have the significance level in comparison with the control group. The ileum experiment in vitro show that Pulsatilla:Coptis=3 : 1,1 : 1 group and the single Pulsatilla have the significance level in comparison with the control group. Conclusion The classical prescriptions of Baitouweng Decoction has the best dissolution of active ingredients and the significant inhibition effect on isolated duodenum.
