摘要
以反相 HPL C法测定了大鼠血浆中克林沙星浓度 ,采用 Spherisorb- C1 8柱 ,以达诺沙星作内标 ,离子对溶液 -乙腈 (90∶ 10 )为流动相 ,方法回收率高 ,结果准确。克林沙星在大鼠体内代谢符合一室模型 ,其峰浓度及 AUC与剂量具有线性关系。
The RP HPLC method for determination of clinafloxacin hydrochloride in plasma of rats was established using Spherisorb C 18 (150×4.6 mm, 5 μm) column with mobile phase acetonitrile pH 3.0 ion pair buffer (10∶90, v/v ) at flow rate of 1.0 ml/min. The detector was set at 297 nm. Calibration curve was linear in the concentration range of 0.05~5.0 μg/ml ( r =0.9999). The RSD of within day and between day was less than 9% with average recoveries of 91.7%~100.1%.
