摘要
采用PCR-SSCP方法对寿光鸡CAPN1基因的外显子14、外显子20及其侧翼区序列进行多态性检测,并分析它们的多态性与部分肉质性状的关系。CE14引物扩增的座位在群体发现3个单核苷酸多态性(SNPs)位点:g.13715409 A>G、g.13715387 T>C位于外显子14中,g.13715467C>T位于内含子13中;共检测到AA、AB、AC、BB、BC及CC 6种不同基因型,基因型频率分别为0.238、0.273、0.197、0.095、0.102、0.095,等位基因A、B、C的频率分别为0.473、0.282、0.245。CE20引物扩增的座位在群体发现6个SNPs位点:g.13713224C>A位于外显子20中,g.13713320A>T、g.13713289G>A、g.13713277T>C位于内含子19中,g.13713185T>C、g.13713184G>A位于内含子20中;共检测到AA、AB、AC、BC、AD、BB、CD及DD 8种不同基因型,基因型频率分别为0.122、0.354、0.197、0.143、0.061、0.034、0.048、0.041,等位基因A、B、C、D的频率分别为0.429、0.282、0.194、0.095。最小二乘统计分析结果表明,CE14座位对胸肌pHu、腿肌pHu、肌内脂肪含量、腿肌剪切力及胸肌蒸煮损失存在显著性影响(P<0.05),CE20座位对胸肌pHu及胸肌蒸煮损失存在显著性影响(P<0.05)。
In this study, the polymorphism of CAPN1 gene in Shouguang chicken was detected using PCR-SSCP method and its relationship with some meat quality traits of Shouguang chicken was also analyzed. At CE14 locus, six genotypes of AA, AB, AC, BB, BC, and CC were found and their frequencies were 0. 238,0. 273,0. 197,0. 095,0. 102,and 0. 095,respectively. Frequencies of allele A,B and C were 0. 473,0. 282, and 0. 245, respectively. Three SNPs were revealed by se- quencing,among which one was in the intron 13(g. 13715467 C〉T),and two were in the exon 14 of CAPNI gene(g. 13715409 A〉G and g. 13715387 T〉C). At CE20 locus, eight genotypes of AA,AB, AC, BC, AD, BB, CD, and DD were found and their frequencies were 0. 122, 0. 354, 0. 197,0. 143,0. 061,0. 034/0. 048, and 0. 041, respectively. Frequencies of allele A, B, C, and D were 0. 429,0. 282,0. 194, and 0. 095, respectively. A total of six SNPs were identified at CE20 lo- cus by sequencing,among which three were in the intron 19(g. 13713320 A〉T,g. 13713289 G~〉 A,and g. 13713277 T〉C) ,one was in the exon 20(g. 13713224 C〉A) ,and two were in the intron 20(g. 13713185 T〉C and g. 13713184 G〉A) of CAPN1 gene. Statistical analysis indicated that the genotypes of CE14 locus showed a significant effect on pHuB,pHuL,IMF,SFL,and CLB(P〈 0.05). The genbtypes of CE20 locus showed a significant effect on pHuB and CLB (P〈0.05).
