八棱海棠类Caspase基因cDNA的克隆及序列分析

摘要 以八棱海棠cDNA为模板,采用RT-PCR、巢式PCR、3’RACE、5’RACE技术,获得了类天冬氨酸特异性的半胱氨酸蛋白水解酶(Caspase)基因cDNA 1429bp的全长序列。序列分析表明,Caspase基因cDNA序列编码358个氨基酸,N端含有1个23个氨基酸的信号肽。进化树分析显示八棱海棠与山杏在该基因座位上亲缘关系最近,属于一个分支。 The caspase-like gene cDNA sequence was cloned with cDNA isolated 1429bp full length cDNA sequence of caspase-like gene was obtained by using RACE and 5' RACE techniques. Further analysis on caspase-like gene cDNA rom malus robust RT-PCR, nested indicates that it a rehd. A PCR, 3 ' encoded a peptide containing 358 amino acids, existing a signal peptide of 23 amino acids in the N terminal. Phylogenetic tree analysis shows malus robusta rehd, and apricot in the Caspase-like gene locus have the closest relationship, which belongs to a branch.
