摘要 目的研究生长分化因子5(Growth and differentiation[actor 5,GDF5)在诱导骨髓间充质干细胞(bonemarrow derived mesenchymal stem cells,BMSCs)成软骨分化中的作用。方法通过全骨髓贴壁分离法体外分离、扩增兔骨髓间充质干细胞,利用腺病毒载体将GDF5基因导入骨髓间充质干细胞,通过Safranin-O染色和甲苯胺蓝染色检测硫酸糖胺聚糖和蛋白聚糖、RT-PCR和免疫化学染色检测软骨细胞特异性CollagenⅡ和Aggrecan基因和蛋白的表达,探索GDF5腺病毒对BMSCs成软骨分化的诱导作用。结果 GDF5腺病毒感染的BMSCs细胞外基质糖胺聚糖和蛋白聚糖的含量显著增加,软骨细胞特异性CollagenⅡ和Aggrecan基因和蛋白表达升高。结论本研究结果表明,GDF5能够显著促进脂肪间充质干细胞的成软骨分化。 Objective To study the effects of GDF5 promoting chondrogenesis of BMSCs. Methods BMSCs were isolated and cultured by adherenee screening method, and then were infected with GDF5 by an adenovirus (Ad-GDFS). Glyeosaminoglycan and proteoglycan, the mRNA and protein expression of chondrocyte marker genes collagen II and ag- greean were determined in the BMSCs under different induce conditions. The promoting ehondrogenic effects of GDF5 were explored. Results GDF5 could increase the eontents of extracellular matrix glyeosaminoglyean and proteoglyean, and up-regulate the genomie and proteinie of ehondroeyte marker genes collagen II and aggreean at the meanwhile. Con- elusion The results of this study demonstrated that GDF5 could promote BMSCs chondrogenesis signficantly.
