摘要
目的研究牡丹籽油对高脂血症大鼠的作用,并观察对糖尿病小鼠及小鼠糖耐量的影响。方法高脂饲料喂养SD雄性大鼠建立高脂血症模型,造模同时连续灌胃给药30 d,检测各组大鼠血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)的水平;尾静脉注射四氧嘧啶诱导高血糖小鼠模型,连续灌胃给药30 d,检测小鼠血糖值;正常小鼠连续给药30 d后,灌胃葡萄糖溶液(2.5 g·kg-1),于给糖前及给糖后30、60、120 min分别检测血糖含量变化。结果高脂血症大鼠在灌胃给予牡丹籽油30 d后,低剂量组TC水平显著降低,高剂量组TG水平明显降低,且HDL-C值显著升高;牡丹籽油各剂量组均显著降低糖尿病小鼠的血糖值,且呈现一定的剂量依赖性;糖耐量实验发现,牡丹籽油可降低正常小鼠的血糖水平,在给糖30 min时的降糖作用最为明显。结论牡丹籽油可降低高脂血症大鼠的血脂水平,同时具有降低糖尿病小鼠血糖、改善正常小鼠糖耐量的作用。
Objective To study the effect of peony seed oil on hyperlipidemic rats,and to explore the effect on diabetic mice and glucose tolerance on normal mice.Methods High fat diet was fed for hyperlipidemic model on SD male rats.At the same time,peony seed oil was intragastric administered for 30 days.Serum TC,TG,HDL-C and LDL-C levels were detected.Diabetic rats model was established by caudal vein injection of alloxan.After 30 days of intragastric administration,glucose level in blood was determined.The normal mice were treated with peony seed oil for 30 days.Then glucose dose of 2.5 g.kg-1 was given to mice with intragastric administration.The blood glucose level was detected pre and post 30,60 and 120 min of intragastric administration of glucose solution respectively.Results After treatment of hyperlipidemic rats with peony seed oil for 30 days,TC level in low-dose and TG level in high-dose decreased significantly.HDL-C level in high-dose increased significantly.All the dose of peony seed oil could significantly reduce the blood glucose level of diabetic mice,and showed dose-dependent manner.The experiment of glucose tolerance showed peony seed oil could also reduce the blood glucose level of normal mice,and was most obvious at 30 min after given glucose solution.Conclusion Peony seed oil could reduce blood lipids level in hyperlipidemic rats,and also had the effect of reducing blood glucose level of diabetic mice and improving glucose tolerance of normal mice.
