摘要
目的探讨自体嗅黏膜移植干预脊髓损伤(spinal cord injury,SCI)大鼠脊髓组织内基质金属蛋白酶-2、9(MMP-2、9)的表达与动物神经功能恢复的关系。方法制作SD大鼠半横断脊髓损伤模型,按随机数字表法分为模型对照组和自体嗅黏膜组,每组20只(同时取5只大鼠设为正常对照组)。自体嗅黏膜组在损伤处植入自体嗅黏膜,移植后1、3、7、14d(每时间点5只大鼠)采用BBB评分评价神经功能、免疫组织化学法检测脊髓内MMP-2、9表达。结果 SCI后自体嗅黏膜组BBB评分高于模型对照组(P<0.05)。各时间点SCI大鼠脊髓内MMP-2、9的表达显著升高(正常对照组大鼠未见MMP-2、9的表达)。伤后同一时间点自体嗅黏膜组脊髓内的阳性细胞数少于模型对照组(P<0.05)。结论自体嗅黏膜移植可明显促进神经运动功能恢复情况;此种作用可能与其能够抑制MMP-2、9过度表达有关。
Objective To study the relationship between the expression of MMP-2 and MMP-9 and the recovery of neurological function in rats undergoing autologous transplantation of olfactory mucosa for injured spinal cord.Methods Healthy Sprague-Dawley rats with spinal hemisection were randomly divided into two groups:control group and autologous olfactory mucosa transplantation group,with 20 rats in each group(Five rats were set to the mormal control group).Neurological function was evaluated by the Basso-Beattie-Bresnahan(BBB) scale and MMP-2 and MMP-9 expression in injured spinal cord was detected by immunohistochemistry 1,3,7 and 14 days after transplantation(5 rats per time point).Results After spinal cord injury,the expression of MMP-2 and MMP-9 obviously increased at all time points(Normal control group no MMP-2 and MMP-9 expression).Compared with control group,the BBB scores increased but the number of MMP-9 and MMP-2 positive cells decreased at the same time point in autologous olfactory mucosa transplantation group(P〈0.05).Conclusion Autologous olfactory mucosa transplantation can promote the recovery of neurological function through inhibiting the expression MMP-9 and MMP-2 in injured rat spinal cord.
