一种无标记基因植物表达载体的改造方法 被引量:1

摘要 利用无标记基因表达载体开展转基因植物研究,是获得无标记基因转基因植物、保证转基因生物安全的手段之一。文章报道了一种无标记基因植物转化载体的改造方法,通过AseⅠ单酶切可去除pCAM-BIA1300植物转化载体内潮霉素标记基因完整表达框架。试验应用结果表明,利用此表达载体进行共转化研究,可获得不含标记基因、只含目的基因的转基因植株。此方法可在植物共转化研究中广泛应用。 Usage of the expressional marker-free vector for plant genetic transformation is a method for obtaining the transgenic plant without selective marker gene.Here,we reported a method for construction of the plant expressional vector based on pCAMBIA1300 backbone by cleavage with the single restriction endonuclease AseⅠ.The constructed vector was used for rice co-transformation,and finally the transgenic rice was obtained without marker gene,which indicated that the potential vector application was to obtain the marker-free plant by co-transformation.
