摘要
基因克隆是进行植物基因功能研究的先决条件。随着拟南芥、水稻、杨树等植物全基因组序列的公布,标志着基因组进入后基因组研究时代是分子生物学发展的必然趋势,同时为进一步克隆植物中控制重要性状的主效基因提供强大的基因组数据支撑。目前有较多的基因克隆传统及衍生技术,文章综述了图位克隆、同源克隆、转座子标记法、表达序列标签法、差异表达基因克隆等5种应用比较广泛的基因克隆技术,并就其发展方向和策略作了展望。
Gene cloning is a prerequisite for its functional identification.Along with the release of genomic sequences of many plants such as Arabidopsis thaliana,Populus trichocarpa,Oryza sativa,it is an inevitable trend of molecular biology with the transition from genome to postgenomics,also it will provide powerful support of genomic data for the cloning of major gene related to some important agricultural characteristics.At present,there are some traditional and derived techniques of gene cloning.In this paper,five methods,such as positional cloning,homologous sequence,transposon tagging,expressed sequence tagging,differentially expressed gene cloning were briefly reviewed in order to provide a useful reference for researchers working in this area.
