反相高效液相色谱法测定水果中有机酸含量 被引量:6

Determination of Organic Acids in Fruit by Reversed Phase High-performance Liquid Chromatography

摘要 利用反相高效液相色谱法同时测定苹果、梨、橙子中草酸、酒石酸、乙酸、乳酸4种有机酸的含量。采用Waters2695 HPLC-PDA色谱系统,Waters Xbridge C18(4.6×250mm,5μm)色谱柱,乙腈和0.01mol/L磷酸二氢钠溶液(pH=2.6)作为流动相,流速1.00mL/min,柱温30℃,紫外检测波长214nm的色谱条件下,能够在10分钟内较好地定量分析水果中4种有机酸的含量,本方法回收率为92.17%-103.19%,相对标准偏差为0.23-0.65,各有机酸标准曲线相关系数均在0.9997以上。本方法准确、高效,灵敏度高,适用于水果中有机酸的检测。 The RP-HPLC method was used for simultaneous determination of four kinds of organic acids including oxalic acid,tartaric acid,acetic acid, lactic acid in apples, pears, oranges.Using Waters2695 HPLC-PDA chromatography system,Waters Xbridge C 18 (4.6×250mm, 5gin) coltmm,acetonitrile and 0.0 lmol/L sodium dihydrogen phosphate solution (PH=2.6) were used as the mobile phase at a flow rate 1.00mL/min,column 30℃,UV detection wavelength 214nm chro- matographic conditions, it can effectively analyze 4 kinds of fruit organic acids within 10 minutes. The recovery rate of the method is 92.17%-103.19%, relative standard deviation is 0.23-0.65, and the organic standard curve correlation coef- ficient is higher than 0.9997.The method is accurate,efficient,high sensitivity for the detection of organic acids in fruit.
