摘要
以鸡的FASN基因序列为参照,通过设计和合成引物扩增出鸽的FASN基因片段,应用基因工程技术通过与pMD18-T载体重组,转化E.coliDH5α感受态细胞,蓝白斑法和PCR法双重鉴定筛选出阳性克隆,进行测序,并将测定的序列与genebank中的鸡FASN基因序列进行比对,结果表明,扩增、测定的鸽FASN基因序列片段长1281bp,GC含量42.00%,AT含量58.00%.测序片段为鸽FASN基因序列的最后一个内含子片段.鸽序列与基因库中家鸡同源序列的保守性为60.52%,比鸡同源序列少154个碱基.
PCR primers to amplify Partial FASN Gene of pigeon are designed and synthesized. The gene FASN fragment is gotten by PCR and connected to vector plasmid pMD18-T. The recombinant plasmid was transfered into E. coli DH 5 a cell and is evaluated by BLUE-WHITE select manner and PCR appraisal manner. The result sequenced to recombinant DNA shows that cloned FASN gene sequence of Columba livia is 1281 bp which has the GC content 42.00% and AT content 58.00% respectively. The result of comparison between the homologous sequences of pigeon and chicken shows that their homology is 60. 52%. The nucleotides of pigeon is 154 less than that of chicken.
