摘要
目的 :探索中药配伍比例对中药复方药效成分药动学的影响。方法 :采用反相高效液相色谱法 ,紫外检测。 3P87药代动力学软件拟合药动学模型 ,统计学处理采用t检验法。结果 :川芎∶赤芍 1∶2和 2∶1芎芍制剂给药组芍药苷的药代动力学模型均为一室开放模型 ,川芎∶赤芍 1∶2和 2∶1组的药代学参数分别为Ka =0 .0 0 82±0 .0 0 10和 0 .0 0 88± 0 .0 0 11(min-1) ,Ke=0 .0 0 6 0± 0 .0 0 15和 0 .0 0 71± 0 .0 0 0 8(min-1) ,t1/ 2Ka=86 .10± 12 .38和79.11± 18.6 9(min) ,t1/ 2Ke=119.95± 2 7.2 3和 98.14± 10 .82 (min) ,tpeak=16 9.16± 2 1.81和 149.39± 2 9.5 1(min) ,Cmax=3845 .89± 716 .70和 16 35 .81± 2 30 .95 ( μg·L-1)。两组药动学参数差异无显著统计学意义 (P >0 .0 5 )。结论 :川芎赤芍配伍对芍药苷的药动学特征无显著影响 ,提示中药配伍并不是必然地导致其中某一种成分的药代动力学特征的变化。
Objective: To explore the effects of dispensing ra tio of Chinese herbs on the pharmacokinetic characteristics of effective compone nts. Methods Paeoniflorin (PF) in serum was determined by RP HPLC with UV detector. The concentration time data were fitted using 3P87 Pharmaco kinetic Program, and the pharmacokinetic parameters were compared by t test. Results:The concentration time curves of PF in both groups of Chu anxiong∶Chishao 1∶2 and 2∶1 Xiongshao Pharmaceutics administrated intragastri cally fitted open one compartment model. Parameters of Chuanxiong∶Chishao 1∶2 and 2∶1 groups∶ Ka=0.0082±0.0010 and 0.0088±0.0011(min -1 ),Ke=0.0060± 0.0015 and 0.0071±0.0008(min -1 ), t 1/2Ka =86.10±12.38 and 79.11± 18.69(min), t 1/2Ke =119.95±27.23 and 98.14±10.82(min), t peak = 169.16±21.18 and 149.39±29.51(min), C max = 3845.89±716.70 and 1635.81 ±230.95 (μg·L -1 )respectively. The pharmacokinetic parameters of the two groups had no statistically significant differences ( p >0.05). Concl usion:Different formulae of Chinese herbs do not always result in changes of pharmacokinetic characteristics of some one component. [
