白花蛇舌草注射液对多发性骨髓瘤细胞株增殖的抑制作用 被引量:7

Inhibitory Effect of Hedyotis Diffusa Injection on Multiple Myeloma Cell Proliferation

摘要 目的探讨白花蛇舌草注射液(HDI)对多发性骨髓瘤(MM)细胞株RPMI8226、U266增殖抑制作用及机制。方法将RPMI8226、U266细胞以1×106/ml的浓度接种于含10%胎牛血清RPMI1640培养液中,取对数生长期的细胞进行实验。检测HDI对RPMI8226、U266抑制作用并筛选研究浓度;观察不同浓度HDI对RPMI8226、U266细胞凋亡、细胞周期、细胞因子及细胞株相关基因mRNA表达的影响。结果筛选出0、20、40、60μl/mlHDI作用于RPMI8226、U266细胞株,HDI可抑制RPMI8226、U266增殖,诱导RPMI8226凋亡,且有G1/S期阻滞作用。HDI可下调RPMI8226、U266上清液血管内皮生长因子(VEGF)水平,且呈浓度依赖,同时上调RPMI8226白细胞介素-6(IL-6)水平、下调U266IL-6水平(P<0.05或P<0.01)。经40μl/mlHDI处理后,RPMI8226及U266细胞株相关基因除Bax、Caspase-3外其余基因mRNA表达均有改变(P<0.05或P<0.01)。结论 HDI可抑制MM细胞株RPMI8226、U266增殖,其作用机制可能促进细胞凋亡、阻滞细胞周期及改变细胞因子及细胞株相关基因mRNA表达有关,但途径不尽相同。 Objective To observe the inhibitory effect of Hedyotis Diffusa Injection (HDI) on the proliferation of multiple myeloma (MM) cell lines RPMI 8226 and U266 and its mechanism. Methods The RPMI 8226 and U266 cells of 1×106/ml concentration were seeded into the RPMI 1640 culture solution with 10% fetal bovine serum, 100 U/ml penicillin and 100 μg/ml streptomycin. The cells in logarithmic growth phase were selected to do the experiment. The inhibitory effect of HDI on RPMI 8226 and U266 was detected and the concentration was screened. The effect of HDI with different concentrations on the apoptosis, cell cycle, cytokines and cell lines related gene mRNA of RPMI 8226 and U266 was observed. Results There were 0, 20, 40 and 60 μl/ml concentrations of HDI selected on RPMI 8226 and U266. HDI could inhibit the proliferation of RPMI 8226 and U266 and induce RPMI 8226 apoptosis with G1/S phase arrest. HDI could down-regulate the vascular endothelial growth factor (VEGF) level of RPMI 8226 and U266 supernatant with concentration dependence. HDI could up-regulate the interleukin-6 (IL-6) level of RPMI 8226 and down-regulate the IL-6 level of U266 (P0.05 or P0.01). The expression of RPMI 8226 and U266 cell lines related genes was changed after processed by 40μl/ml HDI except Bax and Caspase-3 (P0.05 or P0.01). Conclusion HDI can inhibit the proliferation of RPMI 8226 and U266 cells, which may be relevant to promoting apoptosis, arresting cell cycle and changing cytokines and expression of cells related gene mRNA with different way.
