摘要
目的:观察蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后迟发性血管痉挛(delayed cerebral vasospasm,DCVS)大鼠Caveolin-1在基底动脉中的表达情况。方法:SD大鼠150只,分为空白组、生理盐水组和SAH组。SAH组、生理盐水组又分为建模后第3、5、7、14天4组。采用枕大池二次注血法建模,观察基底动脉病理改变,检测Caveolin-1蛋白含量变化。结果:(1)动物建模情况:SAH组大鼠各时间点上Bederson评分≥1分,证明DCVS建模成功;(2)血管痉挛情况:SAH后第3天出现明显管腔变小,第7天管腔变小达高峰,中膜平滑肌层增厚最为明显(P<0.05);(3)Caveolin-1情况:SAH组第7天Caveolin-1表达较生理盐水组及空白组显著下降(P<0.05)。结论:基底动脉Caveolin-1的下调可能与SAH大鼠DCVS有关,可能成为SAH诱发DCVS治疗的潜在靶点,为DVCS的治疗提供方向。
Objective To observe the expression of Caveoion-1 in the basilal artery after delayed cerebral vasospasm (DCVS) in rat model of subarachnoid hemorrhage (SAH). Methods One hundred and fifty SD rats were randomly divided into blank group, saline group and SAH group. The SAH group and saline group were divided into three, five, seven and fourteen days after establishing model. SAH model was established by double injection of autologous arterial blood into the cisterna magana, test the changing of Caveoin-1 in the basal artery. Results (1) The situation of SAH model: in SAH group, all animals Bederson neurological severity scores ≥ 1, proving the success of model. (2) The situation of vasospasm: the lumen of basilar artery in the SAH groups lessened gradually and peaked on the 7th days, the thickness of vascular smooth muscle layer of basilar artery significantly increased (P 〈 0.05). (3) The situation of Caveolin-1 : the protein expression of Caveolin-1 by western blot test in the SAH groups decreased to the lowest point on the 7th days, significantly decreased compared with that in the saline groups and blank groups. Conclusion The protein expression of Caveoin-1 is down-regulated in basilal artery after DCVS in rat model of SAH, Caveoin-1 may be a potential inhibitor for DCVS and provide the direction for DCVS treatment.
