摘要
目的 :探索胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)、胰岛素样生长因子结合蛋白-3(in-sulin-like growth factor binding protein-3,IGFBP-3)与胶质瘤发生发展的关系,为提高胶质瘤的早期诊断率提供实验依据。方法:胶质瘤组及正常对照组(健康志愿者),每组各30例,血清标本均用免疫放射分析法测定IGF-1、IGFBP-3浓度。正常脑组织和肿瘤组织,行常规HE切片染色及IGF-1免疫组化染色检查,显微镜下观察免疫组化染色结果。结果:胶质瘤组血清IGF-1水平明显升高,与对照组比较差异有统计学意义(P<0.01),高恶性胶质瘤表达明显高于低恶度胶质瘤。胶质瘤组血清IGFBP-3水平明显升高,与对照组比较差异有统计学意义(P<0.01),但高、低恶度胶质瘤血清IGFBP-3水平则差异无统计学意义(P>0.05)。二组血清IGF-1水平均与血清IGFBP-3水平呈正相关(r=0.594、0.474、0.375,P<0.05)。正常脑组织中IGF-1无表达;人脑胶质瘤组织中IGF-1表达水平明显升高(P<0.05),高恶度胶质瘤表达明显高于低恶度性胶质瘤(P<0.05)。结论:IGF-1和IGFBP-3可能促进了胶质瘤的发生发展,检测血清IGF-1和IGFBP-3水平对提高胶质瘤的早期诊断率有积极的临床意义。
Objective: To determine expression of insulin-like growth factor-1(IGF-1) in tumor tissues and serum IGF-1、insulin-like growth factor binding protein-3(IGFBP-3) levels in glioma patients,and to investigate both relationships.Meanwhile to discuss the exact roles of IGF-1,IGFBP-3 in the development of glioma.So with the theory,we can detect early glioma by detecting serum IGF-1,IGFBP-3 levels.Methods: The experiment was divided into two groups:healthy control group(healthy volunteer) and glioma group,each group including 30 cases.A 4 mL blood sample from each fasting case was drawn in the early morning,and which was separated by centrifugation immediately and was stored under-20 ℃.Serum IGF-1 and IGFBP-3 levels were determined by immunoradiometric assays(IRMA).To take tissue samples in the operation,and which were made in routine HE staining and IGF-1 immunohistochemical staining.Staining results were observed with microscope.Data were analyzed by χ2 test,and the significance level was P〈0.05.Results:(1)Serum IGF-1 levels in glioma were significantly higher than those in healthy control cases(P〈0.01),and increased serum IGF-1 levels correlated with glioma's malignancy.(2) Serum IGFBP-3 levels in glioma were significantly higher than those in healthy control cases(P〈0.01).(3)Serum IGF-1 levels correlated significantly with serum IGFBP-3 levels in two groups(r=0.594,0.474,0.375,P〈0.05).(4)Less expressions of IGF-1 were detected in normal brain tissues,but expressions of IGF-1 in glioma were obviously increased(P〈0.05).Conclusions: IGF-1 and IGFBP-3 might promote the development of glioma.There was meaningful clinical value in determination's serum IGF-1 and IGFBP-3 levels serving as early indicators of glioma.
