摘要
目的 :通过检测肝细胞肝癌(hepatocellular carcinoma,HCC)患者肝癌、癌旁组织及正常肝组织中NF-κB蛋白和PTEN蛋白表达,结合HCC的临床及病理特征,探讨NF-κB和PTEN基因在HCC发生发展中的机制及意义。方法:用免疫组化方法检测NF-κB、PTEN蛋白在62例HCC组织、对应癌旁组织及20例正常肝组织中的表达情况。结果:NF-κB蛋白在HCC组织的表达水平显著高于癌旁组织和正常肝组织(均P<0.05)。PTEN蛋白在HCC组织的表达水平显著低于癌旁组织和正常肝组织(均P<0.05);NF-κB蛋白和PTEN蛋白的表达在HCC患者不同年龄、性别、肿瘤大小、有无肝硬化背景等特征上,差异无统计学意义(P>0.05),在门静脉是否浸润、有无包膜、分化程度高低上,差异有统计学意义(P<0.05)。HCC组织中NF-κB和PTEN的表达呈负相关(r=-0.729,P=0.000)。结论:NF-κB和PTEN可能参与HCC的发生和发展。两者表达呈负相关提示可能通过PTEN下调和NF-κB上调共同参与了HCC的发生发展。
Objective: To explore the mechanism and significance of PTEN and NF-κB gene in the occurrence and development of hepatocellular carcinoma(HCC) by detecting their protein expression of HCC,corresponding paracancerous tissue and normal liver tissue,combined with the clinical and pathological features of hepatocellular carcinoma.Methods: Immunohistochemical technique was adopted to determine PTEN and NF-κB protein expression in 62 cases of HCC,corresponding paracancerous tissue and 20 cases of normal liver tissue.Results: NF-κB protein expression in human HCC was clearly higher than paracancerous tissue and normal liver tissue(both P〈0.05),PTEN protein expression in human HCC was clearly lower than paracancerous tissue and normal liver tissue(both P〈0.05).NF-κB protein and PTEN protein expression in HCC was not statistically correlated with age,sex,the tumor size,cirrhosis background(P〈0.05).While it was significantly correlated with tumor capsule,portal vein invasion,degree of differentiation(P〈0.05).There is negative correlation between NF-κB protein expression and PTEN protein expression in HCC(r=-0.729,P=0.000).Conclusions: The aberrant expressions of PTEN and NF-κB protein in HCC sugguest that they possiblely be involved in the occurrence and development of HCC.The negative correlation between NF-κB protein expression and PTEN protein expression in HCC points out that it is probably throught up-regulation of NF-κB,down-regulation of PTEN to take part in the occurrence and development of HCC.
