干扰素调节因子1在呼吸道合胞病毒感染后气道神经源性炎症的调控作用 被引量:7

摘要 目的通过研究呼吸道合胞病毒(RSV)感染后的SD大鼠气道反应性和神经源性炎症的变化及干扰素调节因子1(IRF-1)的调控作用,探讨IRF-1在调控RSV感染后调控气道神经源性炎症的作用地位。方法对4组SD大鼠(空白对照组、RSV感染组、IRF-1干扰组和IRF-1基因沉寂组)相应处理后进行气道反应性测定,取其左肺行HE染色,右肺行免疫组化检测P物质(SP)及降钙素基因相关肽(CGRP)。结果①RSV感染组与空白对照组比较:气道反应性明显增高(P<0.05),炎性细胞浸润明显增多,SP和CGRP的表达明显增多;②IRF-1干扰组与RSV感染组比较:气道反应性无明显差别(P>0.05),炎性细胞浸润及SP、CGRP的表达无明显差别;③IRF-1基因沉寂组与RSV感染组相比,气道反应性明显降低(P<0.05),炎性细胞浸润及SP、CGRP的表达明显减少。结论 IRF-1在RSV感染引起气道高反应性的过程中可能是通过参与调节神经源性炎症介质的表达来参与调控,IRF-1有望成为哮喘气道神经源性炎症一个新的治疗靶点。 To investigate the modulatory effect of interferon regulatory factor 1(IRF-1) in respiratory syncytial virus(RSV) infection on the airway neurogenic inflammation by studying the changes of airway responsiveness and neurogenic inflammation after RSV infection and IRF-1 Interference.【Methods】The rats were separated into 4 groups(Control group,RSV-infected group,IRF-1-interfered group and IRF-1-gene silence group),after the different disposal,they were tested airway responsiveness and killed.Their left lungs were observed by hematoxylin-eosin staining.Their right lungs were tested the expressions of substance P(SP) and calcitonin gene-related peptide(CGRP) by immunohistochemistry.【Results】Compared with control group,RSV-infected group had an airway hyper-responsiveness,more severe inflammatory cell infiltration,more SP and CGRP expressions.Compared with RSV-infected group,the airway responsiveness,inflammatory cell infiltration,expressions of SP and CGRP of IRF-1-interfered group were not significant change.The airway responsiveness,inflammatory cell infiltration,expressions of SP and CGRP of IRF-1-interfered group were reduced significant.【Conclusions】In RSV infection caused airway hyper-responsiveness,IRF-1 might be involved in by regulating the expression of neurogenic inflammatory mediators.IRF-1 could be a potential therapeutic target for airway neurogenic inflammation in asthma.
