Cloning and sequence analysis of 2 SFB genes in apricot cultivars belonging to the Central Asian ecotype group

摘要 【目的】为了获得杏中亚生态品种群中自交不亲和花粉特异表达的SFB基因全长,探讨SFB基因序列多态性,【方法】以中亚生态品种群‘馒头玉吕克’和‘索格佳娜丽’2个杏品种为材料,利用RT-PCR及RACE技术和DNAMAN软件分析,克隆得到了2个新的SFB基因的全长cDNA,SFB33(HQ148064)和SFB50(HQ148074)。【结果】SFB33全长1 378 bp,SFB50全长1 313 bp,两个基因开放阅读框均为1 131 bp,编码376个氨基酸;与GenBank上已登录的SFB基因核苷酸序列进行Blast比对,SFB33基因与SFB-c(FJ362527)同源性为79%;SFB50基因与SFB23(EU652886)同源性为99%。在两个基因上均找到F-box区域和两个变异区及两个高变区。李亚科SFB氨基酸序列同源性比对显示SFB50与李亚科其他SFB基因亲缘关系较近,而SFB33与李亚科其他SFB基因亲缘关系较远。【结论】杏中亚生态品种群中SFB基因显示明显的序列多态性。 [Objective]The objective of the study is to clone the complete cDNA sequence of SFB and probe into polymorphism in the Central Asian ecotype group apricot. [Method]2 Apricot (Prunus armenia- ca) cultivars 'Mantouyuliike' and 'Suogejianali' belonging to the Central Asian ecotype group were used as experimental materials and Two complete cDNAs of SFB gene (SFB33 and SFB 50) were cloned by RT- PCR and Rapid Amplification of cDNA Ends (RACE) techniques , and analyzed with DNAMAN software. [Result]The results showed that Two complete cDNAs of SFB gene (SFB33 and SFB 50) were cloned. SFB33 (HQ148064) was 1 378 bp and SFB50 (HQ148074) was 1 313 bp in full length, the two genes were a ORF for 1 131 bp, encoding 376 amino acids. Compared with the nucleotide sequences registered in GenBank, the highest homology of SFB33 was 79% with SFB-c (FJ362527) and SFB50 was 99% with SFB23 (EU652886). F-box regions, two variable regions and two hypervariable regions were both found in the two genes. Based on the deduced amino acid sequence alignment in Prunoideae, SFB 50 was closer with other SFB gene, but SFB50 was far from them in Homology tree. [Conclusion]The studies suggested that there were polymorphism in sequences of SFB gene from the Central Asian ecotvpe ~roup aoricot.
