摘要 【目的】为了获得适宜进行草莓果实类胡萝卜素各组分定性定量分析的方法,【方法】以成熟草莓果实为材料,通过对不同提取剂、提取方法的筛选,优化草莓果实类胡萝卜素的提取条件;通过确定色谱条件以及重复性、精密度、回收率试验,建立类胡萝卜素超高效液相色谱法标准分析体系。【结果】以丙酮∶石油醚=1∶2作为提取剂,皂化提取草莓果实中类胡萝卜素,同时采用ACQUITY UPLC BEH C18色谱柱,以乙腈∶甲醇=9∶1为流动相,在450 nm下测定是进行草莓果实类胡萝卜素定性定量分析的最佳方法。成熟草莓中能检测到叶黄素、β-隐黄质、β-胡萝卜素,含量依次为:118.5、12.6、99.8μg.kg-1;检测不到番茄红素;检测可以在5 min内完成,比常规方法节约时间70%左右;方法平均回收率达到93.2%,变异系数为1.32%~1.86%。【结论】提取方法和超高效液相色谱法标准分析体系适合草莓果实类胡萝卜素各组分的定性定量分析。 [Objective]To obtain suitable analysis method for qualitative and quantitative of carotenoids in mature strawberry fruits, [Method]different solvent and method to extract carotenoids were optimized. Then the absorption wavelength, chromatographic conditions as well as repeatability, precision and re- covery tests were determined to establish carotenoids standard analysis method by ultra performance liquid chromatography (UPLC). [Result]The results showed that the best extract solvent for carotenoids in straw- berry fruit was acetone :petroleum=1:2. Then it should be saponified and analyzed used ACQUITY UPLC BEH C 18 column with acetonitrile:methanol=9:1 as mobile phase at 450 nm. Mature strawberry fruits con- tained Xanthophyll (118.5 p,g.kg-1), ^-Carotene (99.8 p,g'kg-1), 13-Cryptoxanthin (12.6 p,g.kg-l), but Lycopene was not detected here by UPLC. The detection results could be received in 5rain, which saved about 70% of the time. The average recovery of this method was 93.2% and relative standard deviations werel.32% to 1.86%.[Conclusion]The carotenoid extraction and standard analysis method by UPLC were suitable for the carotenoid quality and quantity analysis of strawberry fruits.
