成纤维细胞激活蛋白和基质金属蛋白酶1在小鼠肺纤维化中的表达及相关性 被引量:1

FAP and MMP-I expression and relationship between them in mice pulmonary fibrosis

摘要 目的 探讨成纤维细胞激活蛋白(FAP)在小鼠肺纤维化组织中的表达,及其与基质金属蛋白酶1(MMP-1)之间的关系.方法 制备小鼠肺纤维化模型,采用免疫组织化学技术、Western-bolt检测肺组织中FAP、MMP-1和α-SMA的表达.结果 ①博莱霉素-A5盐溶液诱导后,FAP、MMP-1和α-SMA的表达与小鼠肺泡炎和肺纤维化程度呈正相关;FAP与MMP-1的表达呈正相关.②Westernbolt结果显示FAP对照组表达弱阳性,随肺纤维化程度的增强,表达增强;MMP-1、α-SMA随肺纤维化程度的增加表达亦增强.结论 FAP的表达与肺泡炎和肺纤维化程度均呈正相关,FAP与MMP-1可能在肺纤维化基质重塑中起协同作用. Objective To investigate the expression of fibroblast activation protein (FAP) in mice pulmonary fibrosis and the relationship between FAP and MMP-1.Methods The expressions of FAP,α-SMA and MMP-1 were detected with immunohistochemical and Western blot technique in mice pulmonary fibrosis model.Results ① After BLM-A5 induced,the expression of FAP,MMP-1 and α-SMA were strongly correlated with the stage of lung fibrosis.The expression of FAP was positively correlated with MMP-1.② The expression of FAP was correlated with the lung fibrosis,but weakly positive in control group with Western-bolt analysis.The expression of MMP-1,α-SMA was also correlated with the lung fibrosis degree.Conclusions FAP was strongly correlated with the alveolitis and the lung fibrosis degree.FAP and MMP 1 might play a synergistic role in the ECM remodeling of pulmonary fibrosis.
