A hypothesis of the epiphytic relationship between the pectin and the cutin of cuticle layer in cotton fiber was put forward, and four models including the namely layer style, the layer-dispersion style, the arborescence style and the arboreseence-dispersion style were built, respectively. The pectin content of cotton fiber was measured with copper salt dyeing method and ammonium oxalate extraction before and after removal of waxes. The scanning electron microscopy (SEM) and the Fourier transform infrared attenuated total reflectance spectroscopy (FTIR-ATR) analysis indicate that the model layer style provides the best correlation to the experimental results. The eutin in cotton fiber does not contain free pectin, and there is not any interpenetration, superposition and wrapping, but a rather clear interface between the cutin and the pectin. The cutin is on the outer layer, while the pectin is on the inner layer, which glues the waxy substances to the surface of the cellulose or hemicellulose.
