摘要
目的观察四妙勇安汤对动脉粥样硬化(AS)大鼠辅助性T细胞相关细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)表达的影响,探讨其防治AS的免疫机制。方法将纯种SD雄性大白鼠40只随机分为正常对照组、空白模型组、四妙勇安汤组、普罗布考组,每组10只。制备AS模型,各组于造模后第3天开始给药,四妙勇安汤按15.22 g.kg-1给药,普罗布考组给予普罗布考0.16 g.kg-1,正常对照组、空白模型组给予相同体积的0.9%氯化钠溶液。于实验16周末禁食16 h后,麻醉动物,腹主动脉取血5 mL留置2管,分别分离血清,采用酶联免疫吸附测定(ELISA)法检测TNF-α、IL-10的变化;取大鼠胸腹主动脉(主动脉弓头臂干动脉起始处至髂总动脉分叉处)段,经组织匀浆后取上清液,检测TNF-α、IL-10含量。结果与正常对照组比较,空白模型组大鼠血清和胸腹主动脉IL-10水平显著降低(P<0.01),TNF-α水平显著升高(P<0.01);与空白模型组比较,普罗布考组和四妙勇安汤组TNF-α、IL-10水平均差异有统计学意义(P<0.01);与普罗布考组比较,四妙勇安汤组TNF-α水平差异无统计学意义,IL-10水平差异有统计学意义(P<0.01),四妙勇安汤组优于普罗布考组。结论四妙勇安汤防治动脉粥样硬化的机制与免疫调节有关。
Objective To explore the immune mechanisms of prevention and treatment for atherosclerosis (AS) via atherosclerosis T helper cells associated cytokines expression in rats by Simiao yongan decoction. Methods Forty purebred male SD rats were randomly divided into sham operation, untreated model control, Simiao yongan decoction and probucol groups (n= 10 in each). The AS model was established, and oral medications were started three days later,with 15.22 g · kg-1 Simiao yongan decoction,0.16 g · kg-1 probucol, or the same volume of saline to sham-operated and blank model groups,respectively. By the end of the 16th weekend, the animals were anesthetized after fasting for 16 hours. The serum from 5 mL abdominal aortic bloodwithdrawn from each group, and tissue homogenate supernatant from thoracic and abdominal aorta (from aortic arch head of brachiocephalic artery to the common iliac artery bifurcation) were applied for detecting TNF-α and IL-10 via ELISA assay. Results Compared with the sham-operated control group, the thoracoabdominal aortic IL-IO level was significantly decreased (P〈0. 01 ) ,and TNF-α elevated (P〈0.01 ) in the model group. Compared with the untreated model group, significant difference for TNF-α and IL-10 levels was observed in both the probucol and the Simiao yongan groups (P〈0. 01 ), and Simiao yongan decoction was more active than probucol. Conclusion The mechanism by which Simiao yongan decoction prevents and cures atherosclerosisis may be associated with its immune regulation.
