摘要
目的建立同时测定天山花楸叶中槲皮素和山奈素含量的方法。方法色谱柱:Diamonsil C18(250 mm×4.6 mm,5μm,);流动相为甲醇-乙腈(50∶50);流速0.9 mL.min-1;检测波长:360 nm;柱温:40℃。结果天山花楸中槲皮素的含量为48.22 mg.g-1;山奈素含量为20.57 mg.g-1;平均回收率分别为99.72%,98.58%。结论该方法准确,简便,可作为天山花楸叶中槲皮素和山奈素的含量测定方法。
Objective To establish an HPLC assay for quercetin and kaempferol in the leaves of Sorbus tianschanica Ruper. Methods An HPLC-method was developed using Diamonsil Cjs column (250 mm×d. 6 mm, 5 μm)and the mobile phase cotaining methanol- acetonitrile(50 : 50) , and the UV detection was performed at 360 nm. The flow rate was 0.9 mL · min-1 and temperature was set at 40 ℃. Results The quercetin and kaempferol content was 48. :22 mg · g^-1 and 20.57 mg· g^-1 , respectively. The average recovery ( n = 5 ) of the method was 99. 72% and 98. 58% for quercetin and kaempferol, respectively. Conclusion The method is accurate, simple and can be used for detecting quercetin and kaempferol in the leaves of Sorbus tianschanica Ruper.
