头部重点低温对全脑缺血再灌注损伤鼠ICAM-1mRNA表达及PMNL、M_Φ浸润的影响 被引量:3

Influence of selective brain cooling on the expression of intercellular adhesion molecule-1 mRNA and infiltration of polymorphonuclear leukocytes and monocytes/macrophages in rat with globally cerebral ischemia/reperfusion injury

摘要 目的 评估头部重点低温对全脑缺血再灌注 (I/R)鼠细胞间粘附分子 (ICAM 1)mRNA的表达及多形核白细胞 (PMNL)和单核巨噬细胞 (MΦ)浸润的影响。方法 建立全脑四血管夹闭I/R模型。 35只SD大鼠随机分 5组 ,组Ⅰ :非缺血再灌注组 ;组Ⅱ、组Ⅲ、组Ⅳ、组Ⅴ在头部温度 37℃下全脑缺血 30min后 ,分别在头部温度 37℃、35℃、32℃、2 8℃下再灌注 4h。处死大鼠取脑组织 ,评估PMNL、MΦ浸润 ,逆转录PCR(RT PCR)检测ICAM 1mRNA的表达。结果 组Ⅱ与组Ⅰ比较 ,ICAM 1mRNA表达及PMNL、MΦ 浸润显著增加 (P <0 0 1) ;与组Ⅱ比较 ,组Ⅲ、组Ⅳ及组Ⅴ的ICAM 1mRNA表达及PMNL、MΦ 浸润明显降低 (P <0 0 5或 0 0 1)。结论 I/R损伤可诱导ICAM 1mRNA表达和PMNL、MΦ浸润 ;头部重点低温 ,特别是中度低温 (2 8℃~ 32℃ )能显著抑制ICAM 1mRNA表达 ,减轻PMNL和MΦ在脑的浸润 ,产生更好的脑保护作用。 Objective To evaluate effects of selective brain cooling on the expression of intercellular adhesion molecule 1(ICAM 1)mRNA and infiltrations of polymorphonuclear leukocytes(PMNL) and monocytes/macrophages(M Φ) during global cerebral ischemia/reperfusion (I/R) Methods Global ischemia of brain was produced by the four vessel occlusion for 30 min followed by 4 h reperfusion Thirty five SD rats were randomized into five groups: group I without ischemia and reperfusion; group II,group III,group IV or group V with cerebral ischemia at 37 degrees centigrade followed by reperfusion at 37,35,32 or 28 degrees centigrade respectively The cerebral tissues samples were taken for pathologic examination of the infiltration of PMNL and M Φ and to detect the ICAM 1mRNA expression by reverse transcription PCR (RT PCR) Results The expression of ICAM 1mRNA and infiltration of PMNL and M Φ increased remarkably in the group II,group III,group IV and group V compared with those in group I (P<0 01); As compared with those in group II,The expression of ICAM 1mRNA and infiltration of PMNL and M Φ decreased evidently (P<0 05 or 0 01) Conclusions I/R injurys induce ICAM 1mRNA expression and PMNL and M Φ infiltrations; selective brain cooling, especially moderate hypothermia (28 32 degrees centigrade), provides better cerebral protection by means of inhibiting the expression of ICAM 1mRNA and the infiltrations of PMNL and M Φ in brain
