摘要
目的 :筛选与分离枸杞子中抑制人前列腺细胞增殖的活性成分。方法 :水、95 %乙醇、5 0 %乙醇、丙酮与正丁醇等溶剂分别提取 ,经硅胶柱色谱和TLC分离 ;MS ,IR ,1HNMR ,UV确定结构。MTT显色试验测定提取物的抑制人前列腺癌PC3 细胞增殖活性。结果 :从枸杞子 5 0 %乙醇提取物中分得单体化合物莨菪亭 ,并证明有显著的抑制PC3 细胞增殖活性。结论 :莨菪亭是枸杞子抑制PC3 细胞增殖的活性成分。
Objective:To extract and isolate the active component inhibiting PC 3 cell proliferation in vitro from the fruit of Lycium barbarum .Method:The active components of the fruit of L. barbarum were extracted by water,95% ethanol,50% ethanol respectively. Then, ethanol extract was extracted with acetone and n butanol and isolated by SiO 2 column and purified by TLC to obtain the active component. The chemical structure of active component was identified by the spectral analysis. MTT assay was used to compare the inhibition activity(IC 50 ) of PC 3 cells proliferation of the all extracts.Result:According IC 50 ,the most active component among all extracts isolated from the fruit of L. barbarum isscopoletin.Conclusion:Scopoletin is the active component of the fruit of L. barbarum for inhibiting PC 3 cell proliferation.
