人脐带间充质干细胞对大鼠烧伤的治疗作用 被引量:3

Therapeutic effect of UC-MSCs on burned rats

摘要 目的探讨人脐带间充质干细胞(UC-MSCs)对重度烧伤大鼠的治疗作用。方法分离纯化人UC-MSCs。24只SD大鼠均分为两组:治疗组烧伤后实施UC-MSCs移植;对照组烧伤后滴入PRMI 1640培养液。HE染色观察大鼠皮损的病理结构,比较创面愈合率。结果成功培养获得UC-MSCs,所得细胞分别经形态学、流式细胞术、体外分化方法作表型鉴定。治疗组大鼠创面愈合时间为(22.0±3.1)d,短于对照组的(27.0±3.1)d(P<0.05)。术后14d,治疗组创面愈合率为(56.3±9.8)%,高于对照组的(47.2±8.3)%(P<0.05)。结论局部UC-MSCs移植能有效促进烧伤组织恢复。 Objective To investigate the efficacy of umbilical core mesenchymal stem cells(UC-MSCs)in treatment of burn in rats.Methods UC-MSCs were isolated and identified in vitro.Twenty-four burned rats were egually randomized into two groups of A(locally transplanted with UC-MSCs seeding)and B(dropped with PRMI 1640culture solution as control).The pathological structure of wounded tissues was observed by HE staining.The wound healing rate was compared.Rusults UC-MSCs were obtained successfully and identified by morphology,flow cytometry and cell differentiation.The wound healing time was shorter in group A than that in group B[(22.0±3.1)d vs.(27.0±3.1)d](P〈0.05).On the 14th day after treatment,the wound healing rate was higher in group A than that in group B [(56.3±9.8)% vs.(47.2±8.3)%](P〈0.05).Conclusion Local transplantation of UC-MSCs can promote the skin wound healing in rats with burn injury.
