栀子苷改善CCl_4引起的小鼠肝纤维化损伤的实验研究 被引量:18

摘要 目的研究栀子苷的抗肝纤维化药理作用。方法雄性C57小鼠采用腹腔注射CCl4构建肝纤维化模型,同时采用栀子苷进行灌胃给药干预。运用HE染色和天狼星红染色检测肝纤维化的变化;生化检测血清丙氨酸转移酶(ALT)和天冬氨酸转移酶(AST)以及肝组织的羟脯氨酸含量。Western blot法检测转化生长因子β1(TGF-β1)及α-SMA的蛋白表达。结果栀子苷显著抑制CCl4诱导的小鼠肝纤维化程度。与纤维化模型组相比,栀子苷治疗组血清ALT、AST水平及肝脏组织中羟脯氨酸水平明显降低(P<0.01)。栀子苷显著抑制肝组织中TGF-β1和α-SMA的蛋白表达(P<0.01)。结论栀子苷具有显著的改善肝纤维化作用。 Objective To investigate the anti-hepatofibrotic effects of geniposide on liver fibrosis. Methods Male C57 mice were induced with 20% CCl4(2 mL/kg) for 6 weeks to induce liver fibrosis, and were treated with intragastrical garage of 50 mg/kg geniposide for 14 d since the fifth week of CC14 induction. Liver fibrosis of the mice was evaluated by HE staining and Sirius Red staining. Activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and liver content of hydroxyproline were measured with automated biochemistry analyzer. The protein levels of transforming growth factor betal ( TGF-β1 ) and α- smooth muscle actin (ot-SMA) were detected by Western blot analysis. Results Geniposide significantly at- tenuates the progression of liver fibrosis induced by CC14. Treatment with geniposide also resulted in significant decreases in the serum levels of ALT and AST, as well as the content of hydroxyproline after CCI4 induction(P 〈 0. 01 ). Geniposide significantly reduced the protein expression levels of TGF-β1 and α-SMA in the liver(P〈0.01.Conclusin Geniposide exerts anti-fibrotic effects on the liver induced by CCl4.
