汉族和裕国族人群GP-340基因多态性与龋病易感性的相关性

摘要 目的:研究唾液糖蛋白GP-340基因rs1969619(T/G)、rs1969620(A/G)、rs2684742(C/T)三个位点单核苷酸多态性(SNPs)与裕固族和汉族人群龋病易感性的关系。方法:采用病例对照研究,从甘肃裕固族自治县常住人口中选取两民族样本355例,其中病例组(DMFT≥3)164例(汉族85例,裕固族79例),对照组(DMFT≤2)191例(汉族105例,裕固族86例);分别采集各受试者外周静脉血并提取DNA;经PCR反应后,其产物用核酸质谱技术(MassArray)检测GP-340基因中3个位点的基因型;结果用SPSS 16.0软件包进行统计分析。结果:所有基因型分布频率均符合Hardy-Weinberg平衡(P>0.05);GP-340基因3个多态性位点基因型频率,在裕固族病例组和对照组中的分布均无统计学差异(P>0.05);而在汉族人群中,rs1969619位点GT基因型的频率分布,病例组为32.9%,对照组为55.2%[P<0.05,OR=0.392,95%CI=0.197~0.779]。结论:GP-340基因rs1969619位点多态性可能与汉族人群龋病易感性有关。 AIM: To investigate the relationship between the three SNPs of GP-340 gene and dental caries susceptibility in the population of Han and Yugur nationality. METHODS: 355 individuals in Yugur autonomous counties of Gansu province were enrolled in a case-control study. Case group( DMFT≥3 ) included 164 individuals(85 of HaM nationality and 79 of Yugur) ,control group(DMFT≤2) 191 ( 105 of HaM hationality and 86 of Yugur). DNA was extracted from peripheral venous blood. The distribution of the genotype frequency of s1969619(T/G), rs1969620 (A/G) and rs2684742 (C/T)was examined by Sequenom MassArray. The data were statistically analyzed by SPSS 16.0 software package. RESULTS: All genotypes were in Hardy-Weinberg equilibrium range(P 〉0.05). In Yugur individuals the frequencies of the 3 genotypes were not significantly different between case and control groups (P 〉0.05). In HaM individuals rs1969619 SNP genotype frequencies in case and control groups were 32.9% and 55.2% respectively (P 〈 0.05, OR = 0. 392,95% C1 = 0. 197 - 0. 779). CONCLUSION : The genetic polymorphism in the locus of rs1969619 of GP - 340 gene might be associated with dental caries susceptibility in HaM population.
