葛根素对庆大霉素致大鼠肾毒性的保护作用 被引量:4

摘要 目的:研究葛根素(Puerarin,Pue)对庆大霉素大鼠肾毒性的保护作用。方法:雄性SD大鼠给予溶媒或葛根素(50 mg.kg-1.d-1)7 d后,再联合给予0.9%氯化钠注射液或庆大霉素140 mg/kg(i.p.)14 d,测定大鼠体重,血清尿素氮(BUN)、血肌酐(SCr)含量,计算肌酐清除率(CCr),测定肾组织中丙二醛(MDA)和一氧化氮(NO)含量和超氧化物歧化酶(SOD)及内皮一氧化氮合酶(eNOS)活性;HE染色观察肾脏病理改变。结果:与正常对照组比较,庆大霉素组可致大鼠体重、CCr、NO和SOD活性下降,血清BUN和SCr及肾组织MDA含量升高,肾脏病理改变明显。给予葛根素明显减轻上述改变。结论:葛根素有效拮抗庆大霉素所致的大鼠肾损害,可能通过其抗氧化作用及提高NO水平来发挥作用。 AIM: To explore the possible pro- tective effects of puerarin (Pue) against genta- micin (GM)-induced nephrotoxicity in rats. METHODS: Male rats were i. p. administrated of Pue (50 mg. kg-1 .d-1) or vehicle (0. 9% NaCl) for 7 days before and 14 days concurrently with saline or GM (140 mg /kg, i.p. ). Body weight, serum levels of creatinine (SCr) and blood ureanitrogen (BUN) and creatinine clear- ance rate (CCr) were assessed, the level of ma- londialdehyde (MDA), nitric oxide (NO), the activities of superoxide dismutase (SOD) and en- dothelial nitric oxide synthase (eNOS) in renal tissue were determined. The renal histopatho- logical changes were observed by hematoxylin-e- osin staining. RESULTS. Compared with thecontrol group, rats treated with GM showed de- crease in body weight, Ccr, NO and the SOD ac tivity in kidney, and a significant elevation of se- rum SCr, BUN and the renal MDA contents and severe morphology changes. Pue significantly reversed the changes mentioned above. CON- CLUSION: Pue provide marked protective effect against GM-induced nephrotoxicity in rats, which may be related to their antagonistic effects on oxidative stress, and the increment of NO level.
