破壁方法对葡糖醋杆菌TDY1破壁率及乙醇脱氢酶酶活的影响

Effect of different cell wall disruption methods on cell wall disruption rate and alcohol dehydrogenase activity in Gluconocetobacter oxydans TDY1 cell

摘要 分别用超声破碎法、反复冻融法、溶菌酶法和细菌裂解液法等4种不同破壁方法对葡糖醋杆菌TDY1进行破壁处理,通过比较细胞的破碎率和乙醇脱氢酶的活力,确定适合于菌株TDY1的细胞破壁方法和最佳条件。结果表明,超声破碎法获得的乙醇脱氢酶活力最高、为4 004(±224)U/mg,破壁率为97.3(±1.26)%,与其他3种方法比较差异极显著;处理2 mL菌数为1.3×1010CFU/mL的菌液时,超声破碎的最佳条件为:超声9 s、破碎总时间75 s、功率95 W。 Four different cell wall disruption methods, including ultrasonic treatment, repeated freezing, lysozyme digestion and cell lysis solution were used to treat Gluconocetobacter oxydans TDY1 ceils respectively. By emnparing the cell wall disruption rate and the activities of alcohol dehydrogenase, the papaer chose the most appropriate method for cell wall disruption of TDYI. The results showed that ultrasonic treatment collected the highest activity of alcohol dehydrogenase with 4 004 (±224) U/rag, and the cell wall disruption rate was 97.3 (±1.26)%, significantly different from the others. When authors disrupted 2 mL cell suspension with 1.3 ×10^10 CFU/mL by ultrasonic treatment, the optimum conditions were ultrasonic time 9 s, breaking time 75 s, ultrasonic power 95 W.
