摘要 目的:分离培养人羊水干细胞(hAFSC)并鉴定。方法:使用贴壁法分离培养羊水干细胞,细胞免疫荧光法和Western blot鉴定hAFSC。结果:分离的羊水干细胞均表达特异性标记物Oct-4、c-kit、SSEA-4、CD105。结论:成功分离及培养hAFSC,可作为种子细胞用于干细胞移植治疗肝纤维化。 Objective: To isolate,culture and identify the stem cells derived form human amniotic fluid(hAFSC).Methods: hAFSCs were isolated and cultured with adherent method,and identified by cell immune fluorescence and Western blotting.Results: Generally,the isolated amniotic stem cells expressed specific markers of Oct-4,c-kit,SSEA-4,and CD105.Conclusion: hAFSC is successfully isolated and cultured,which can be used as the seed cells for management of hepatic fibrosis through stem cell transplantation.
