环孢素A血药浓度的HPLC测定方法

摘要 目的:建立环孢素A血药浓度的HPLC测定方法。方法:采用反相HPLC的方法,血样经乙醚萃取,正己烷洗脱等处理后测定。采用DiamonsilTMC18色谱柱,流动相为乙腈-水(74:26,V/V),流速为1ml/min,柱温为70℃,检测波长为210nm。结果:环孢素A血药浓度在50~1000ng/ml范围内线性关系良好,R2=0.9997,低中高三种浓度的提取回收率为70.01%~75.00%,方法回收率为98.93%~103.08%,日内精密度RSD为0.84%~4.76%,日间精密度RSD为5.38%~9.87%。结论:该方法快速灵敏准确,可用于环孢素A血药浓度的监测。 Objective: To establish a HPLC method of detecting cyclosporine A concentration in blood. Method: Cyclosporine A was extracted from whole blood with diethyl ether, and the endogenous interference was removed from blood sample with n-hexane. HPLC was adopted including with DiamonsilTM C18 column and the 70 ~C temperature.The mobile phase was consisted of acetonitrile-water ( 74: 26, V/V ) at the flow rate of 1 ml/min with the detection wavelength at 210 nm. Result: The linear range of cyclosporine A was good ( R2=0.9997 ) .The extraction recovery was 70.01%-75.00% and the method recovery was 98.93%-103.08%.The intra-day precision RSD was 0.84%-4.76%, and the inter-day precision RSD was 5.38%~9.87%. Conclusion: The established method is rapid, sensitive and accurate, therefore, it is suitable for the drug monitoring of cyclosporine A in the clinic.
