摘要
本文运用 DIG标记的人源小卫星探针 33.6与 HaeⅢ消化的中华绒螯蟹基因组 DNA杂交,建立了中华绒螯蟹非放射性 DNA指纹图谱,证明目前在缺乏来自中华绒螯蟹基因组 DNA本身的同源小卫星 DNA的情况下,运用异源小卫星探针进行中华绒螯蟹 DNA指纹图谱的研究是可行的;实验表明运用人源小卫星探针 33.6进行中华绒螯蟹 DNA指纹图谱研究时,该探针可检测到 10个以上位点的遗传变异,每个位点呈现丰富的多态性。
The studides established non- radioactive DNA fingerpringting of Chinese mitten- handed crabs(Eriocheir sinensis) by hybridizing HaeⅢ completely dogested genomic DNA of Chinese mitten- handed crabs with DIG labeled probe of human mini- satellite 33.6.It showed that studying on DNA fingetpringting of Chinese mitten- handed crabs with non- homologous mini- satellite probes was feasible in short of homologous mini- satellite probes cloned from gDNA of Chinese mitten- handed crabs.DNA fingerpringting of Chinese mitten- handed crabs produced by human mini- satellite 33.6 detected over 10 locus in gDNA of Chinese mitten- handed crabs and those locus showed abundence of polymorphism.
