尿激酶型纤溶酶原激活物介导的肿瘤靶向性重组人肿瘤坏死因子-α的生物活性 被引量:2

摘要 目的利用先前制备好的尿激酶型纤溶酶原激活物(uPA)介导的肿瘤靶向性重组人肿瘤坏死因子-α(rhTNF-α)融合蛋白,验证uPA对融合蛋白的去封闭效果,并对融合蛋白的生物学活性进行初步研究。方法以hTNF-α标准品为参照,对切除GST标签的融合蛋白(即foldon-uPA1-hTNF-α)和经uPA酶切后的去封闭融合蛋白(即rhTNF),用MTT法检测融合蛋白的细胞毒性、Hoechst 33258/PI荧光染色检测融合蛋白对CNE2-Z细胞的作用。结果 uPA对融合蛋白具有去封闭效果,融合蛋白和去封闭融合蛋白能诱导高表达uPA的肿瘤细胞凋亡。结论经过改造的uPA介导的肿瘤靶向性rhTNF-α融合蛋白具有高效低毒和靶向性,为TNF的临床应用提供了理论基础。
