摘要
目的筛选利福平依赖结核杆菌,并对其分子生物学特征进行初步鉴定。方法用含利福平(200μg/mL)的酸性罗氏培养基分离培养疑似结核病人的痰液,对筛选出菌株的rpoB基因进行测序分析。结果筛选出5株利福平依赖菌株,测序结果表明所有菌株在rpoB的核心突变区内均有突变发生。结论本研究筛选的利福平依赖菌株在ropB核心突变区内均有突变发生,但仍需进一步研究利福平依赖菌株的其他分子生物学特征。
Objective To identify rifampicin -dependent Mycobacterium tuberculosis,and preliminary analysis its molecular characteristics. Method The sputum specimens from suspected TB patients was isolated and cultured on acidic L-J medium containing 200 p~ g/ml rifampein, and 81-bp rifampin resistance-determining region (RRDR) sequences of rpoB gene were determined for screened strains. Results We have screened five rifampicin-dependent strains, and all of the five isolates had mutantions in the RRDR of the M. tuberculosis rpoB gene. Conclusion In this study, all of the rifampicin-dependent strains have mutantions in the RRDR of rpoB gene, but further studies will be required to define other molecular characteristics of these strains.
