摘要
目的:建立人血草干燥全草中黄连碱和白屈菜红碱含量的测定方法.方法:采用反相HPLC法.色谱柱为YMC-Pack ODS-AA(4.6mm×250mm,5μm),以乙腈-水(0.2%磷酸+0.2%三乙胺)体系梯度洗脱,检测波长280nm,流速1.0mL.min-1,柱温30℃.结果:黄连碱在13.24~264.8μg.mL-1质量浓度范围内面积与浓度呈良好的线性关系(r=0.999 7);加样回收率为99.4%,RSD为1.1%(n=6).白屈菜红碱在6.36~127.2μg.mL-1质量浓度范围内面积与浓度呈良好的线性关系(r=0.999 8);加样回收率为101.6%,RSD为1.2%(n=6).结论:本法简单、准确,重复性好,可用于人血草药材的质量.
This paper develops a method for determining of the content of coptisine and chelerythrine in Stylo- phorum lasiocarpum (Oliv.) Fedde. A HPLC method is established using YMC-Pack ODS-AA(4.6 mm X 250 mm, 5 t_tm) with the mobile phase acetonitrile and water (0.2 % phosphoric acid q- 0.2 % triethylamine) in gradient mode. The detection wavelength is set 280 nm. The flow rate is 1.0 mL ~ rain-~ and column temper- ature is set at 30~C. The linear relationship of coptisine is determined with the range from 13.24--264.8~g ~ mL-a (r=0. 999 7). The average recovery as 99.4% with RSD 1.1%. The linear relationship of chelerythrine is determined with the range from 6.36--127.2/~g ~ mL-~ (r=0. 999 8). The average recovery as 101.6% with RSD 1.2 %. The method is simple, accurate and reliable and can be used for quality control of Stylopho- rum lasiocarpum (Oliv.) Fedde.
