白细胞介素10对大鼠腹腔感染和早期脓毒症炎症抑制作用的实验研究 被引量:6

摘要 目的 利用动物实验方法探讨白细胞介素 10 (interleukine 10 ,IL 10 )在腹腔感染和脓毒症时有无抑制炎症的作用。方法  48只SD大鼠随机分为 4组 ,每组 12只。第 1组单纯腹腔切开 ;第 2组腹腔切开后行盲肠结扎和穿刺 (cecalligationandpunctures,CLP) ;第 3组 ,术前 1h腹腔内注入IL 10 15 0 0 0U ,CLP术后再每 3h分别注入IL 10 15 0 0 0U ;第 4组 ,术前不用药 ,CLP术后每 3h分别每次注入IL 10 15 0 0 0U。各组大鼠分 2次在术后 3h和 9h处死 ,取下腔静脉血测炎性细胞因子水平及肝、肺、肠组织作病理切片检查。结果 CLP能使大鼠血中炎性细胞因子水平升高 ,IL 10注入腹腔降低了早期脓毒症时血中这些炎性细胞因子水平 ,并在一定程度上减轻了内脏组织的炎症改变。结论 IL 10对大鼠腹腔感染和早期脓毒症时的炎症有一定的抑制作用 ,提示它可能是有效的抗炎性物质之一。 Objective To explore the effect of IL 10 on inflammatory inhibition of intraabdominal infection and sepsis in mice models. Methods Forty eight SD mice were divided equally into 4 groups namely Group 1 underwent laparotomy; Group 2 received cecal ligation plus punctures(CLP) in which normal saline was injected once every 3 hrs; Group 3 injected CLP and IL 10 intraperitoneally and Group 4 IL 10 was injected 1 hr before and every 3 hrs following CLP. The four groups were equally divided into two batches and sacrificed at 3 and 9hr after CLP. The blood cytokines were measured , and the liver, lung and intestine specimen were examined microscopically . Results The cytokines were not detected in group 1, while TNF α (tumor necrosis α), IL 1 and IL 6(interleukine 1,6) in the serum of the other 3 groups were increased by CLP. IL 10 caused the cytokines decreasing to some degree and attenuated the visceral inflammatory reaction. Conclusion IL 10 could inhibit the release of cytokine and suppress inflammatory reaction in mice models. It may act as an anti inflammation modulator.
