摘要
在骨损伤愈合过程中,血管形成和骨再生紧密相连。血管系统不仅为骨组织的发育和再生提供营养物质和氧气,还能传送促进间充质细胞向成骨细胞分化的主要信号分子。低氧诱导因子(HIFs)和低氧模仿剂(HMAs)被认为是促进血管形成的主要刺激物。低氧环境中,HIFs能促进血管形成-骨再生,而HIFs在常氧中会快速降解。HMAs主要通过在常氧条件下抑制HIFs的降解,从而达到促进血管形成-骨再生。本文对低氧对血管形成-骨再生分子机制的影响做一综述。
Angiogenesis and osteogenesis is a coup-ling phenomenon during bone healing. The vascular system supplys nutrients and oxygen to the developing and regenerating bone, and delivers critical signals which stimulate mesenchymal cell differentiation towards an osteogenic phenotype. Hypoxia inducible factors (HIFs) and hypoxia-mimicking agents (HMAs) are considered to be key stimulators of blood vessel formation. HIFs can triggers angiogenesis and osteogenesis under hypoxia. However, HIFs rapidly degrade under normoxia. HMAs triggersangio-genesis and osteogenesis by preventing degradation of HIFs under normoxic condition. In this article, we review the current studies of the cellular and molecular mechanisms regulating angiogenesis and osteogenesis.
