琼脂糖微囊化成猪胰岛异种移植实验研究

摘要 目的 观察实验性糖尿病大鼠腹腔内移植琼脂糖微囊化成猪胰岛治疗效果。方法 以琼脂糖作为制备微囊的材料 ,用相分离方法包被成猪胰岛 ,将 18只糖尿病大鼠随机分为 3组 :对照组、未微囊化胰岛移植组、微囊化胰岛移植组 ,分别进行腹腔内胰岛移植。结果 移植前 3组血糖水平无显著差异 ,移植后 7天时 ,3组血糖分别为 (2 2 .6 7± 1.15 ) mmol/ L,(18.5 8± 2 .6 2 ) mm ol/ L 和 (12 .38± 2 .6 8) m mol/ L,微囊化胰岛移植组与前 2组相比均有显著差异 ,且生存期明显长于前 2组。结论 琼脂糖微囊化成猪胰岛可存活于糖尿病大鼠体内 ,并且具有生物活性。 Objective To observed the effect of intraperitoneal microencapsulated adult pig islets with agarose gel in experimental diabetic rats. Methods The microencapsules were made by phase separation method with agarose as an immunoisolation membrane. The eighteen diabetic rats induced by streptozotocin were separated into three groups: control group, unencapsulated islets transplantation group and encapsulated islets transplantation group. The unencapsulated and encapsulated pig islets were respective put into peritoneal cavity of diabetic rats. Results There were no significant difference in blood glucose before transplantation among the three groups. On the 7th day after transplantation, the mean levels of the blood glucose in the three groups were (22.67±1.15) mmol/L, (18.58±2.68) mmol/L and (12.38±2.68) mmol/L, respectively. There is significant difference between the encapsulated islets transplantation group and other two groups ( P <0.01, P <0.05, respectively). The encapsulated islets transplantation group survived much longer than the other two groups. Conclusion Microencapsulated adult pig islets with agarose may promote survival rate in diabetic rats.
