Determination of Cordycepin in Fermentation Supernatant of Cordyceps militaris by HPLC and LC-MS/MS: A Comparison of Two Methods

摘要 目的:通过高效液相色谱(HPLC)与液质联用(LC-MS/MS)测定蛹虫草发酵液中虫草素的方法比对,评价2种方法的可行性。方法:蛹虫草发酵液经大孔树脂分离纯化后,分别采用紫外检测器和质谱检测器对10种不同发酵液进行测定。结果:HPLC法测定发酵液中虫草素的检出限(LOD)和定量限(LOQ)分别为13.6μg/L和53.6μg/L,LC-MS/MS法测得虫草素的LOD和LOQ分别为2.1μg/L和7.8μg/L,2种方法虫草素的保留时间分别为22.7min和7.8min,平均回收率分别为94.68%和93.91%。结论:两种方法灵敏度高、选择性好、操作简便,均可对蛹虫草发酵液中虫草素进行准确的定性定量分析。 Objective: To comparatively evaluate the feasibility of HPLC and LC-MS/MS for measuring cordycepin in fermentation supernatant of Cordyceps militaris.Methods: The fermentation supernatants of 10 Cordyceps militaris strains were purified by macroporous resin column chromatography.The purified samples obtained were analyzed separately by HPLC with UV detection and LC-MS/MS.Results: The limit of detection(LOD) and limit of quantitation(LOQ) of HPLC were 13.6 μg/L and 53.6 μg/L,respectively,whereas those of LC-MS/MS were 2.1 μg/L and 7.8 μg/L,respectively.The retention time of cordycepin in HPLC and LC-MS/MS was 22.7 min and 7.8 min,respectively,and the average recovery was 94.68% and 93.91%,respectively.Conclusion: Both methods have high sensitivity,selectivity and simple operation,and can be used for qualitative and quantitative analysis.
