钙敏感受体在颈动脉平滑肌细胞增殖和迁移中的作用 被引量:4

摘要 目的观察钙敏感受体(calcium-sensing receptor,CaSR)在大鼠颈动脉平滑肌细胞中的表达情况及在颈动脉平滑肌细胞增殖和迁移中的作用。方法通过Realtime-PCR和Western Blot检测颈动脉平滑肌细胞中CaSR的表达情况;通过MTT法和Transwell法观察CaSR对颈动脉平滑肌细胞增殖和迁移能力的影响;通过Western Blot检测CaSR对MMP-9、MMP-2和PCNA表达的影响。结果 RT-PCR和Western blot结果显示在颈动脉平滑肌细胞中有CaSR的表达;MTT法和Transwell法结果显示与正常对照组相比较,CaSR激动剂组颈动脉平滑肌细胞增殖和迁移能力明显增强(P<0.05),而CaSR抑制剂组颈动脉平滑肌细胞增殖和迁移能力明显降低(P<0.05);Western Blot检测发现与正常对照组相比较,CaSR激动剂组MMP-9、MMP-2和PCNA表达明显增强,而CaSR抑制剂组MMP-9、MMP-2和PCNA表达明显降低。结论颈动脉平滑肌细胞中有CaSR的表达,并且CaSR可以影响颈动脉平滑肌细胞增殖和迁移。
