摘要 目的:探讨肿瘤坏死因子-α(TNF-α)对血管内皮细胞ICAM-1表达的诱导作用。方法:通过血管内皮细胞培养,采用免疫组织化学方法分别观察不同浓度(0~250U·ml-1)TNF-α24小时内及某一浓度(100U·ml-1)TNF-α在不同时程(0~72h)对内皮细胞ICAM-1表达的影响。结果:与对照组比较,TNF-α呈浓度与时间依赖性诱导内皮细胞ICAM-1的表达。结论:TNF-α是诱导血管内皮细胞表达ICAM-1的直接原因之一。 Objective: Regulation of the Intercellular Adhesion Molecule-1 (ICAM-1) expression by Tumor Necrosis Factor-α (TNF-α) in Vascular Endothelial Cells was investigated. Methods: Human Umbilial Vein Endothelial Cells (HUVEC) were stimulated with TNF-α at the indicated concentration (0 -250U · ml-1) for the same time (24h) and with the same concentration (100U · ml-1) for the different time (0-72h), cell surface expression of ICAM-1 was detected by immunocytochemistry methods. Results; After the exposure of endothelial cells to TNF-α, the expression of ICAM-1 in endothelial cells was obviously increased compared to control group (P<0. 05 or 0. 01), the induction was concentration and time-dependent. Conclusion: TNF-α may directly induce the expression of ICAM-1 in Vascular Endothelial Cells.
